He did have the vampiric strength, you're not wrong on that. He just didn't have all of it yet and grew stronger as the fight progressed. He only turned fully around the time Polnareff exposed him to light since he wasn't aware of the effect sunlight would have on him in that moment.

Vanilla Ice becomes a vampire in his dying moments because Dio gave him some of his blood. It's the entire reason he even died in that fight—he disintegrated in the sun.
It wasn't sarcasm. It genuinely would probably kill him. I can't think of another way calamity could kill him besides internal damage like a stroke, appendicitis, or others. If we're talking human Vanilla Ice than 100%, but he died seconds before becoming a full vampire. Vampire Vanilla Ice might be impenetrable though.

For some reason my idiotic theory with cheap trick is that he wouldn't see her and thus couldn't get her, but I'm now also realizing nobody could ever actually see her back because she's invisible.

Also, Pucci doesn't need 4km or anything. He just needs to survive for 20 to 30 seconds so he can outrun Dio long enough for time to accelerate.
King Crimson erases time and existence overall for its duration, with the exception of Diavolo who gets to move outside of it as normal. He uses epitaph to predict the location someone will be, it doesn't; however, give him super human reflexes. If that person is passing through at 12,000km per hour he can predict when they'll be in a certain location but he can't just automate the time the he exits time erasure to the exact billionth of a second the person shows up inside of his fist. This is also assuming time acceleration doesn't just absolutely $h1t on the usage of his stand as he sees 10 seconds of normal time as opposed to 10 seconds of accelerated time or erase 10 seconds of accelerated time as opposed to normal time (in which both circumstances Pucci will be moving too fast to see, or not moving at all)
Both of these are based on reaction speed. Dio needs to react fast enough to Diavolo, and Diavolo has to warn Dio in the exact .000001seconds that Pucci is present.
Pucci's speed increases exponentially up to infinity for all of 5 minutes. He could just wait 40km away for the final seconds earth remains then annihilate them since by that time since Dio's going to have to pause it on the exact plenk time [5.39 × 10^(-44) seconds] that Pucci moves in. It's a one in a very multitudinous number of a gamble, and Dio doesn't have plot armour.
